Abstract

There are significant antigenic differences between A/equi‐2 (H3N8) strains Miami/ 63, Stockholm/68, and Solvalla/79 when they are compared with antisera raised in ferrets. Both the H and N antigens differ. Two additional strains, Borås/79 and France/79 are closely related to Solvalla/79. Sera from horses vaccinated with the Miami/63 or Stockholm/68 strain distinguish the vaccine strains from the Solvalla/79 isolate as regards the H antigen. The same horse sera did not show differences between the N antigens as the ferret sera do. For titration of antibodies to the N antigen, the lectin‐NI test proved to be reliable. The strains have also been compared by polyacrylamide gel electrophoresis of their polypeptides and genomic RNA segments. For all the strains, the polypeptide pattern was similar except for the Stockholm/68 strain which had a slower mobility of the HA‐2 polypeptide. Differences in the size of RNA segments were demonstrated for isolates from separate outbreaks. The 1979 isolates could be distinguished from the strains of the 1963 and 1968 outbreaks on the basis of their RNA pattern.
Influenza equi 2: RNS‐ und Proteinmuster in Variäntstammen Signifikante Antigenunterschiede existieren zwischen den A/equi‐2 (H3N8)‐Stämmen Miami/63, Stockholm/68 und Solvalla/79 beim Vergleich mit Frettchenserum. Sowohl H‐ als auch N‐Antigene differieren. Zwei weitere Stämme Borås/79 und Frankreich/79 sind eng mit Solvalla/79 verwandt. Seren von Pferden, die mit den Stämmen Miami/63 oder Stockholm/68 geimpft wurden, unterscheiden die Impfvirusstämme vom Solvalla/79‐Isolat im H‐Antigen. Die Pferdeseren zeigten die mit Frettchenseren nachgewiesenen Unterschiede bei den N‐Antigenen nicht. Für die Titration von N‐Antikörpern war der Lectin‐N‐Hemmungstest geeignet. Ferner wurden in der PAGE die RNS‐ und Polypeptidmuster der Stämme verglichen. Polypeptidmuster waren bei allen Stämmen gleich, mit Ausnahme des Stammes Stockholm/68, der ein langsamer wanderndes H‐2 Polypeptid aufwies. Bei den RNS‐Segmenten ergaben sich Unterschiede bei Isolaten von verschiedenen Ausbrüchen. Die Isolate von 1979 konnten aufgrund der RNS‐Muster leicht von den in den Jahren 1963 und 1968 gewonnenen Virusisolaten unterschieden werden.

No sólo diferían los antígenos H sino también los N. Otras dos estirpes, Borås/79 y Francia/79, tienen un parentesco estrecho con Solvalla/79. Los sueros sanguíneos de caballos que fueron vacunados con las estirpes Miami/63 o Estocolmo/68 diferencian las estirpes de virus vacunal del aislamiento Solvalla/79 en el antígeno H. Los sueros de caballo no mostraron las diferencias puestas en evidencia en los antígenos N con los sueros de hurones. Para la titulación de anticuerpos N resultaba apropiada la prueba de inhibición lectina‐N. Además se compararon en la PAGE los modelos de ARN y polipéptidos de las estirpes. Los modelos de polipéptidos eran similares en todas las estirpes, excepción hecha con la estirpe Estocolmo/69, la cual presentaba un polipéptido H‐2 de migración más lenta. En los segmentos ARN se infirieron diferencias en los aislamientos de diversos brotes. Los aislamientos logrados en 1979 se pudieron diferenciar con suma facilidad de los aislamientos de virus obtenidos en los años 1963 y 1968 en virtud de los modelos de ARN.

<p>There are significant antigenic differences between A/equi‐2 (H3N8) strains Miami/ 63, Stockholm/68, and Solvalla/79 when they are compared with antisera raised in ferrets. Both the H and N antigens differ. Two additional strains, Borås/79 and France/79 are closely related to Solvalla/79. Sera from horses vaccinated with the Miami/63 or Stockholm/68 strain distinguish the vaccine strains from the Solvalla/79 isolate as regards the H antigen. The same horse sera did not show differences between the N antigens as the ferret sera do. For titration of antibodies to the N antigen, the lectin‐NI test proved to be reliable. The strains have also been compared by polyacrylamide gel electrophoresis of their polypeptides and genomic RNA segments. For all the strains, the polypeptide pattern was similar except for the Stockholm/68 strain which had a slower mobility of the HA‐2 polypeptide. Differences in the size of RNA segments were demonstrated for isolates from separate outbreaks. The 1979 isolates could be distinguished from the strains of the 1963 and 1968 outbreaks on the basis of their RNA pattern.</p>

<p>Signifikante Antigenunterschiede existieren zwischen den A/equi‐2 (H3N8)‐Stämmen Miami/63, Stockholm/68 und Solvalla/79 beim Vergleich mit Frettchenserum. Sowohl H‐ als auch N‐Antigene differieren. Zwei weitere Stämme Borås/79 und Frankreich/79 sind eng mit Solvalla/79 verwandt. Seren von Pferden, die mit den Stämmen Miami/63 oder Stockholm/68 geimpft wurden, unterscheiden die Impfvirusstämme vom Solvalla/79‐Isolat im H‐Antigen. Die Pferdeseren zeigten die mit Frettchenseren nachgewiesenen Unterschiede bei den N‐Antigenen nicht. Für die Titration von N‐Antikörpern war der Lectin‐N‐Hemmungstest geeignet. Ferner wurden in der PAGE die RNS‐ und Polypeptidmuster der Stämme verglichen. Polypeptidmuster waren bei allen Stämmen gleich, mit Ausnahme des Stammes Stockholm/68, der ein langsamer wanderndes H‐2 Polypeptid aufwies. Bei den RNS‐Segmenten ergaben sich Unterschiede bei Isolaten von verschiedenen Ausbrüchen. Die Isolate von 1979 konnten aufgrund der RNS‐Muster leicht von den in den Jahren 1963 und 1968 gewonnenen Virusisolaten unterschieden werden.</p>

<p>Des différences antigéniques significatives existent entre les souches A/equi‐2 (H3N8) Miami/63, Stockholm/68 et Solvalla/79 en comparant avec des immunsérums de furets. Aussi bien les antigènes H que N sont diffirents. Deux autres souches, Borås/79 et Francel/79 sont étroitement apparentées h Solvalla/79. Les sérums de chevaux inoculés avec les souches Miami/63 ou Stockholm/68 se différencient de la souche vaccinale de l'isolement Solvalla/79 dans l'antigène H. Les sérums de chevaux n'ont pas montré les différences avec les antigènes N mis en évidence avec les sérums de furets. Le test d'inhibition de N à la lectine fut approprié pour la titration des anticorps N. De plus, des échantillons d'ARN et de polypeptides des souches ont été comparés dans PAGE. Les échantillons de polypeptides furent identiques pour toutes les souches, à l'exception de la souche Stockholm/68 qui présenta une migration plus lente du polypeptide H‐2. Il y a eu des différences dans les segments d'ARN avec des isolements de différentes extractions. <p>Diferencias antigénicas significantes existen entre las estirpes A/equi‐2 (H3N8) Miam/63, Estocolmo/68 y Solvalla/79 en comparación con sueros inmunitarios de hurones. No sólo diferían los antígenos H sino también los N. Otras dos estirpes, Borås/79 y Francia/79, tienen un parentesco estrecho con Solvalla/79. Los sueros sanguíneos de caballos que fueron vacunados con las estirpes Miami/63 o Estocolmo/68 diferencian las estirpes de virus vacunal del aislamiento Solvalla/79 en el antígeno H. Los sueros de caballo no mostraron las diferencias puestas en evidencia en los antígenos N con los sueros de hurones. Para la titulación de anticuerpos N resultaba apropiada la prueba de inhibición lectina‐N. Además se compararon en la PAGE los modelos de ARN y polipéptidos de las estirpes. Los modelos de polipéptidos eran similares en todas las estirpes, excepción hecha con la estirpe Estocolmo/69, la cual presentaba un polipéptido H‐2 de migración más lenta. En los segmentos ARN se infirieron diferencias en los aislamientos de diversos brotes. Los aislamientos logrados en 1979 se pudieron diferenciar con suma facilidad de los aislamientos de virus obtenidos en los años 1963 y 1968 en virtud de los modelos de ARN.</p>

Die Pferdeseren zeigten die mit Frettchenseren nachgewiesenen Unterschiede bei den N‐Antigenen nicht. Für die Titration von N‐Antikörpern war der Lectin‐N‐Hemmungstest geeignet. Ferner wurden in der PAGE die RNS‐ und Polypeptidmuster der Stämme verglichen. Polypeptidmuster waren bei allen Stämmen gleich, mit Ausnahme des Stammes Stockholm/68, der ein langsamer wanderndes H‐2 Polypeptid aufwies. Bei den RNS‐Segmenten ergaben sich Unterschiede bei Isolaten von verschiedenen Ausbrüchen. Die Isolate von 1979 konnten aufgrund der RNS‐Muster leicht von den in den Jahren 1963 und 1968 gewonnenen Virusisolaten unterschieden werden.</abstract>
Influenza equi 2: échantillon d'ARN et de protéine dans des souches variants Des différences antigéniques significatives existent entre les souches A/equi‐2 (H3N8) Miami/63, Stockholm/68 et Solvalla/79 en comparant avec des immunsérums de furets. Aussi bien les antigènes H que N sont diffirents. Deux autres souches, Borås/79 et Francel/79 sont étroitement apparentées h Solvalla/79. Les sérums de chevaux inoculés avec les souches Miami/63 ou Stockholm/68 se différencient de la souche vaccinale de l'isolement Solvalla/79 dans l'antigène H. Les sérums de chevaux n'ont pas montré les différences avec les antigènes N mis en évidence avec les sérums de furets. Le test d'inhibition de N à la lectine fut approprié pour la titration des anticorps N. De plus, des échantillons d'ARN et de polypeptides des souches ont été comparés dans PAGE. Les échantillons de polypeptides furent identiques pour toutes les souches, à l'exception de la souche Stockholm/68 qui présenta une migration plus lente du polypeptide H‐2. Il y a eu des différences dans les segments d'ARN avec des isolements de différentes extractions. Les isolements de 1979 ont pu être facilement différenciés des isolements des virus obtenus en 1963 et 1968 sur la base de l'échantillon d'ARN.
<abstract lang="es">Influenza equi 2: Modelos de ARN y proteínas en estirpes variants Diferencias antigénicas significantes existen entre las estirpes A/equi‐2 (H3N8) Miam/63, Estocolmo/68 y Solvalla/79 en comparación con sueros inmunitarios de hurones. No sólo diferían los antígenos H sino también los N. Otras dos estirpes, Borås/79 y Francia/79, tienen un parentesco estrecho con Solvalla/79. Los sueros sanguíneos de caballos que fueron vacunados con las estirpes Miami/63 o Estocolmo/68 diferencian las estirpes de virus vacunal del aislamiento Solvalla/79 en el antígeno H. Los sueros de caballo no mostraron las diferencias puestas en evidencia en los antígenos N con los sueros de hurones. Para la titulación de anticuerpos N resultaba apropiada la prueba de inhibición lectina‐N. Además se compararon en la PAGE los modelos de ARN y polipéptidos de las estirpes. Los modelos de polipéptidos eran similares en todas las estirpes, excepción hecha con la estirpe Estocolmo/69, la cual presentaba un polipéptido H‐2 de migración más lenta. En los segmentos ARN se infirieron diferencias en los aislamientos de diversos brotes. Los aislamientos logrados en 1979 se pudieron diferenciar con suma facilidad de los aislamientos de virus obtenidos en los años 1963 y 1968 en virtud de los modelos de ARN.</abstract>
