Abstract
Abstract: Setting: Chloroquine, an alkalinizing lysosomotropic agent, enhances the intracellular activity of antibiotics against Mycobacterium tuberculosis or Coxiella burnetii.Objective: To determine if chloroquine modifies the activity of clarithromycin, less effective at acidic pH, against intracellular Mycobacterium avium.Design: The activity of clarithromycin (4 μg/ml) against the MO-1 strain of M. avium was evaluated within human macrophages in presence of chloroquine (5 μg/ml). The minimal inhibitory concentration of clarithromycin for the strain was 2 μg/ml.Results: While clarithromycin alone did decrease the intracellular infection at day 7 of culture (P < 0.01), chloroquine alone did not impede the intracellular growth of M. avium, and did not enhance the activity of clarithromycin.Conclusion: Chloroquine should not improve clarithromycin treatment against M. avium infection.
Résumé: Cadre: La chloroquine, agent alcalinisant lysosomotrope, augmente l'activité des antibiotiques contre Mycobacterium tuberculosis ou Coxiella burnetii.Objet: Déterminer si la chloroquine modifie l'activité de la clarithromycine, moins active à pH acide, contre Mycobacterium avium intracellulaire.Schéma: L'activité de la clarithromycine (4 μg/ml) contre la souche MO-1 de M. avium a été évaluée dans des macrophages humains en présence de chloroquine (5 μg/ml). La concentration minimale inhibitrice de la clarithromycine pour la souche était de 2 μg/ml.Résultats: Tandis que la clarithromycine seule a diminué l'infection intracellulaire au jour 7 de culture (P < 0,01), la chloroquine seule n'a pas empêché la croissance intracellulaire de M. avium, et n'a pas augmenté l'activité de la clarithromycine.Conclusion: La chloroquine ne renforce pas le traitement par clarithromycine de l'infection à M. avium.
