Abstract

Abstract: Current methods of tissue culture are described, including techniques for the measurement of cell viability and growth. Literature on the utilization of tissue culture to measure the toxicity of a wide range of substances is reviewed. Of a variety of techniques used, most depend on reduction of viability or growth rates, but inhibition of cell respiration, morphological change and several other parameters have also been observed. However, there appears to have been no systematic comparison of the reaction of normal adult human tissues and animal tissues in toxicological studies. This paper offers proposals for research in which a selection of animal tissues would be compared with human tissue in their response to substances for which substantial data from in vivo toxicology already exist. The results of such work should permit a relatively simple predictive test procedure to be selected.

<abstract lang="en">Abstract: Current methods of tissue culture are described, including techniques for the measurement of cell viability and growth. Literature on the utilization of tissue culture to measure the toxicity of a wide range of substances is reviewed. Of a variety of techniques used, most depend on reduction of viability or growth rates, but inhibition of cell respiration, morphological change and several other parameters have also been observed. However, there appears to have been no systematic comparison of the reaction of normal adult human tissues and animal tissues in toxicological studies. This paper offers proposals for research in which a selection of animal tissues would be compared with human tissue in their response to substances for which substantial data from in vivo toxicology already exist. The results of such work should permit a relatively simple predictive test procedure to be selected.</abstract>
