Abstract
ABSTRACT The usability of fish species identification with restriction fragment length polymorphism (RFLP) using high‐performance liquid chromatography (HPLC) was evaluated for sturgeon, salmon, and tuna samples. Separations of restriction fragments were performed within about 10 min at alkaline pH using an anion‐ex‐change diethylaminoethyl nonporous column. Unequivocal species identification was achieved with HPLC, despite nucleotide sequence‐depending separation. Separation of DNA fragments by HPLC could be demonstrated to be a fast and reliable alternative to electrophoresis. Calculation of restriction fragment lengths using relative peak areas showed better accordance compared with values obtained by means of DNA size standards.
