Abstract
La maladie de Parkinson (MP) est une affection neurodégénérative fréquente, d'étiologie inconnue. Depuis longtemps, l'existence de neurotoxines affectant de manière préférentielle la voie dopaminergique nigro-striatale laisse entrevoir la possibilité que des facteurs environnementaux jouent un rôle dans sa pathogenèse. Au cours de ces dernières années, la découverte de gènes responsables de rares syndromes parkinsoniens familiaux a apporté un nouvel éclairage aux mécanismes dégénératifs en jeu dans la MP. Un nombre croissant d'études montre que les produits de certains de ces gènes interagissent avec des toxines environnementales et interviennent dans des voies moléculaires régulant l'intégrité fonctionnelle de la mitochondrie.
