Abstract

Résumé: Les virus présentent une diversité génétique supérieure aux autres organismes. L'absence d'activité de correction associée aux polymérases virales ARN dépendantes engendre de nombreuses mutations. L'organisation génomique de certaines familles virales permet de plus le phénomène de réassortiment génétique et/ou de recombinaison.La variabilité génétique est reliée d'une part au type d'infection induite par le virus (aiguë ou persistante) et d'autre part aux pressions de sélections exercées par le système immunitaire de l'hôte infecté. Les mécanismes mutationnels concernent aussi bien les régions codantes (protéines d'adhésion du virus grippal, du VIH1…), que des régions impliquées dans la régulation de l'expression des gènes viraux (région amorce de la traduction du poliovirus…). L'exemple de l'HTLV montre au contraire que certains virus possèdent une extraordinaire stabilité provenant de l'absence de multiplication virale au sein de la cellule ayant intégré le génome viral.Cette grande capacité d'adaptation et d'évolution des virus a des répercussions importantes en terme de pathogénicité, de thérapie et de prévention.
Summary: Virus are characterized by a high genetic flexibility rarely found in other microorganism. The high error rate of polymerases (RNA polymerase and reverse transcriptase) is known to generate a great number of mutations, and in some viral families, genetic variability could arise from exchange of genetic markers (recombination, reassortment).Other factors may contribute to viral diversity in individual infection. In acute infection, mutation rate is low, due to the rapid clearance of the virus and the viral divergence appears over time after transmission between individuals or between patient to animal (i.e Influenza). In persistent infection with a high rate of replication (i.e HIV) virus remaining in patient for life, the divergence of viral strains is shaped by a combination of mutations and selection forces. In persistent infection with absence of viral replication (i.e HTLV) virus harbor a high genetic stability.Mutation events occur in coding region (i.e hemagglutinin of Influenza virus) or non coding region (i.e Poliovirus internal ribosome entry site).Genetic variability occurring during viral infection will have consequential effects on viral disease progression, on therapeutic strategy and on viral disease control.

Les mécanismes mutationnels concernent aussi bien les régions codantes (protéines d'adhésion du virus grippal, du VIH1…), que des régions impliquées dans la régulation de l'expression des gènes viraux (région amorce de la traduction du poliovirus…). L'exemple de l'HTLV montre au contraire que certains virus possèdent une extraordinaire stabilité provenant de l'absence de multiplication virale au sein de la cellule ayant intégré le génome viral.</ce:simple-para>

<ce:simple-para>Virus are characterized by a high genetic flexibility rarely found in other microorganism. The high error rate of polymerases (RNA polymerase and reverse transcriptase) is known to generate a great number of mutations, and in some viral families, genetic variability could arise from exchange of genetic markers (recombination, reassortment).</ce:simple-para>
<ce:simple-para>Other factors may contribute to viral diversity in individual infection. In acute infection, mutation rate is low, due to the rapid clearance of the virus and the viral divergence appears over time after transmission between individuals or between patient to animal (i.e Influenza). In persistent infection with a high rate of replication (i.e HIV) virus remaining in patient for life, the divergence of viral strains is shaped by a combination of mutations and selection forces. In persistent infection with absence of viral replication (i.e HTLV) virus harbor a high genetic stability.</ce:simple-para>
<ce:simple-para>Mutation events occur in coding region (i.e hemagglutinin of Influenza virus) or non coding region (i.e Poliovirus internal ribosome entry site).</ce:simple-para>
<ce:simple-para>Genetic variability occurring during viral infection will have consequential effects on viral disease progression, on therapeutic strategy and on viral disease control.</ce:simple-para>

<abstract lang="fr">Résumé: Les virus présentent une diversité génétique supérieure aux autres organismes. L'absence d'activité de correction associée aux polymérases virales ARN dépendantes engendre de nombreuses mutations. L'organisation génomique de certaines familles virales permet de plus le phénomène de réassortiment génétique et/ou de recombinaison.La variabilité génétique est reliée d'une part au type d'infection induite par le virus (aiguë ou persistante) et d'autre part aux pressions de sélections exercées par le système immunitaire de l'hôte infecté. Les mécanismes mutationnels concernent aussi bien les régions codantes (protéines d'adhésion du virus grippal, du VIH1…), que des régions impliquées dans la régulation de l'expression des gènes viraux (région amorce de la traduction du poliovirus…). L'exemple de l'HTLV montre au contraire que certains virus possèdent une extraordinaire stabilité provenant de l'absence de multiplication virale au sein de la cellule ayant intégré le génome viral.Cette grande capacité d'adaptation et d'évolution des virus a des répercussions importantes en terme de pathogénicité, de thérapie et de prévention.</abstract>
<abstract lang="en">Summary: Virus are characterized by a high genetic flexibility rarely found in other microorganism. The high error rate of polymerases (RNA polymerase and reverse transcriptase) is known to generate a great number of mutations, and in some viral families, genetic variability could arise from exchange of genetic markers (recombination, reassortment).Other factors may contribute to viral diversity in individual infection. In acute infection, mutation rate is low, due to the rapid clearance of the virus and the viral divergence appears over time after transmission between individuals or between patient to animal (i.e Influenza). In persistent infection with a high rate of replication (i.e HIV) virus remaining in patient for life, the divergence of viral strains is shaped by a combination of mutations and selection forces. In persistent infection with absence of viral replication (i.e HTLV) virus harbor a high genetic stability.Mutation events occur in coding region (i.e hemagglutinin of Influenza virus) or non coding region (i.e Poliovirus internal ribosome entry site).Genetic variability occurring during viral infection will have consequential effects on viral disease progression, on therapeutic strategy and on viral disease control.</abstract>
