Abstract

Emblem of bioorganometallics, ferroquine is the most advanced organometallic drug candidate and about to complete phase II clinical trials as a treatment for uncomplicated malaria. This ferrocene-containing compound is active against both susceptible and resistant Plasmodium falciparum and Plasmodium vivax strains and/or isolates to the most used drug, chloroquine. This article sums up the discovery of ferroquine, its antimalarial activity, the hypothesis of its mode of action and the current absence of resistance in vitro.

<p>Emblème de la bioorganométallique, la ferroquine est le candidat médicament organométallique le plus avancé dans son développement industriel et devrait achever des études cliniques de phase II comme traitement du paludisme non compliqué. Ce composé à base de ferrocène est extrêmement actif contre des isolats et/ou des clones de Plasmodium falciparum sensibles ou résistants à l’antipaludique le plus utilisé, la chloroquine, et contre Plasmodium vivax. Cet article résume la découverte de la ferroquine, son activité antipaludique, les hypothèses liées à son mode d’action et l’absence actuelle de résistance in vitro.</p>
