Data of the fluorescence signal (F) were obtained during the melting transition of the fluorescein-labeled detection probe from the amplified fragment. Fluorescence data were converted to derivative melting curves by plotting the negative derivative of the fluorescence with respect to temperature against temperature (−(dF/dT) vs. T). The derivative melting curve is plotted for a sample homozygous for wild-type allele (A), a heterozygous sample (B), and a sample homozygous for the mutant allele (C). Melting analysis of a no template control (D) was also performed.</note>
