Abstract

Incubation of mouse embryo cells with 5-fluorouracil and thymidine does not prevent the multiplication of pseudorabies virus, but does affect the multiplication of polyoma virus. The degree of inhibition of the growth of polyoma virus is dependent upon the length of time prior to infection that the cultures are exposed to the drug. This inhibition is not due to a direct effect of 5-fluorouracil on viral messenger RNA; it occurs even when 5-fluorouracil is not available in the intracellular pool for incorporation into viral messenger RNA. Inhibition by the drug of the multiplication of polyoma virus is an “all or none” phenomenon. While 5-fluorouracil suppresses completely in some of the cells all aspects of the infective process that were tested, in other cells in the drug-treated cultures infection appears to proceed at a normal rate, indicating that an early step in the infective process is sensitive to 5-fluorouracil. The nature of this early event is discussed.
