Abstract

Rabbit anti-rat sera have been prepared with rat thymocytes, rat thoracic duct lymphocytes, and rat spleen lymphocytes. Anti-lymphocyte globulins were extracted from the antisera. The ability of the antisera and their globulins to induce the take, and promote the growth of a Walker sarcoma tumor challenge in rats immune to the tumor, was studied, as well as their effect on peripheral blood lymphocytes. The toxicity of the products was defined by the mortality accompanying the treatment. Thymocytes and thoracic duct lymphocytes raised more potent and less toxic ALS than did spleen lymphocytes. The preferential role of thymus-derived over bone-marrow-derived lymphocytes, relative to the specificity, effectiveness, toxicity, and induced peripheral blood lymphopenia, of heterologous ALS is discussed. The second-set tumor allograft is proposed as a sensitive and convenient model to detect the differences of a small degree between immunosuppressive products.
