Cytochrome b sequence divergence in the European sea bass (Dicentrarchus labvax) and phylogenetic relationships among some Perciformes species

Abstract

The entire cytochrome b (cyt b) gene has been sequenced in eight Mediterranean populations of the European sea bass, Dicentrarchus labrax and in one of D. punctatus. Our data indicate that both eastern (Greece and Egypt) and northwestern (French coast) populations of D. labrax were genetically differentiated from the Tyrrhenian ones, which were nearly indistinguishable from each other at the cyt b level. The D. labrax population from the Atlantic coast (Portugal) was genetically quite distinct from all the Mediterranean ones. These results strongly confirm the conclusions from previous studies where the same populations were screened for allozymes, random amplified polymorphic DNA (RAPD) and mitochondrial DNA (mtDNA) d‐loop variation. Seven other species of Perciformes belonging to five different families (Sparidae, Serranidae, Carangidae, Pomatomidae, Sciaenidae) were sequenced to explore the usefulness of the cyt b gene for inferring evolutionary relationships at different hierarchical levels. The data were analysed together with other published cyt b sequenecs from Perciformes fishes. Our data suggest that the superfamily Percoidea is not monophyletic. At the family level, the Sparidae and Moronidae seem to be monophyletic. The evolutionary relationships among families were not resolved. Possible causes for this lack of resolution are discussed.

<p>Sequenzdivergenz im Cylochrom b des Europäischen Meerbarsches (Dicentrarchus labrax) und die phylogenetischen Beziehungen zwischen einigen Arten der Barschartigen Das gesamte Cytochrom b‐Gen (cyt b) wurde für acht verschiedene mediterrane Populationen des Europäischen Meerbarsches (Dicentrarchus labrax) und für eine Population von D. punctatus sequenziert. Unsere Ergebnisse weisen darauf hin, daß sowohl die östlichen (Griechenland und Ägypten) als auch die nordwestlichen (französische Küste) Populationen von D. labrax von den tyrrhenischen Populationen, die untereinander auf der Ebene des cyt b nicht unterscheidbar sind, genetisch differenziert sind. Auch eine Population der Atlantikküste (Portugal) war deutlich verschieden von den mediterranen Populationen. Diese Beobachtungen bestärken die früheren Resultate, die bei Analysen mit Allozym‐, RAPD‐ und mtDNA‐D‐Loop‐Varianten gewonnen wurden, nachhaltig. Die cyt b‐Gene von sieben weiteren Arten der Perciformes, die fünf verschiedenen Familien (Sparidae, Serranidae, Carangidae, Pomatomidae, Sciaenidae) angehören, wurden ebenfalls sequenziert, um zu überprufen, ob die cyt b‐Sequenzen auch für die Aufdeckung phylogenetischer Beziehungen auf verschiedenen hierarchischen Ebenen brauchbar sind. Die Daten wurden zusammen mit anderen, bereits früher publizierten cyt b‐Sequenzen von barschartigen Fischen verglichen. Unsere Ergebnisse weisen darauf hin, daß auf der Ebene der Familien die Sparidae und die Moronidae monophyletisch zu sein scheinen. Die evolutionären Prozesse innehalb von Familien konnten nicht erkannt werden. Die moglichen Ursachen für diese Unsicherheiten werden diskutiert.</p>

<p>The entire cytochrome
<i>b</i>
(cyt
<i>b</i>
) gene has been sequenced in eight Mediterranean populations of the European sea bass,
<i>Dicentrarchus labrax</i>
and in one of
<i>D. punctatus.</i>
Our data indicate that both eastern (Greece and Egypt) and northwestern (French coast) populations of
<i>D. labrax</i>
were genetically differentiated from the Tyrrhenian ones, which were nearly indistinguishable from each other at the
<i>cyt b</i>
level. The
<i>D. labrax</i>
population from the Atlantic coast (Portugal) was genetically quite distinct from all the Mediterranean ones. These results strongly confirm the conclusions from previous studies where the same populations were screened for allozymes, random amplified polymorphic DNA (RAPD) and mitochondrial DNA (mtDNA)
<sc>d</sc>
‐loop variation. Seven other species of Perciformes belonging to five different families (Sparidae, Serranidae, Carangidae, Pomatomidae, Sciaenidae) were sequenced to explore the usefulness of the
<i>cyt b</i>
gene for inferring evolutionary relationships at different hierarchical levels. The data were analysed together with other published cyt
<i>b</i>
sequenecs from Perciformes fishes. Our data suggest that the superfamily Percoidea is not monophyletic. At the family level, the Sparidae and Moronidae seem to be monophyletic. The evolutionary relationships among families were not resolved. Possible causes for this lack of resolution are discussed.</p>
